Potential new HIV vaccine/therapy target

ScienceDaily (May 30, 2012) ? After being infected with simian immunodeficiency virus (SIV) in a laboratory study, rhesus macaques that had more of a certain type of immune cell in their gut than others had much lower levels of the virus in their blood, and for six months after infection were better able to control the virus.

SIV is a retrovirus that infects primates. Strains of SIV that crossed over to humans resulted in the evolution of HIV. In rhesus macaques, SIV causes simian AIDS (though in many primates it is harmless) and studying the virus in these animals offers crucial insights into how HIV acts in humans, the researchers said.

The discovery by researchers at UCSF may shed light on the mystery of why some people infected with HIV are better able to control the virus, live longer and have fewer associated health problems than others who have been infected as long, they said. It also provides a potential new target for developing therapies or vaccines.

The cells that have the protective effect, called Th17 (T helper 17) cells, are a subset of the type of disease-fighting immune cell targeted and killed by HIV and found in the gut of both primates and humans.

A prior study from the same UCSF team found that SIV infection causes a normally protective immune response to infection to go awry, leading to reduction in the protective activity in the gut of these Th17 cells and weakening of mucosal defenses against bacteria. Interestingly, in that study, Th17 cells were not affected by SIV in another primate, African green monkeys, in which SIV infection is harmless and does not cause disease.

"Animals with more of these Th17 cells were better able to control SIV and this was due in part to macaques developing a more effective immune response by producing more SIV-specific CD4-positive T-cells to fight the infection. Our next step is to see if we can augment the Th17 effect, perhaps by looking at interleukin 17 (IL-17), the cytokine released by these cells, and testing to see if it has an effect," said the study's primary investigator, Dennis Hartigan-O'Connor, MD, PhD, assistant professor of medicine at the UCSF Division of Experimental Medicine.

"Further, if a treatment can be developed to increase Th17 cells in the gut, it may allow for a more effective immune response after exposure to an HIV vaccine or the virus itself," he added.

The findings are being published in the May 30, 2012 issue of Science Translational Medicine.

In the new study, the investigators first determined the levels of Th17 cells in the gut of sixteen rhesus macaques and then infected them with SIV. They found that the animals with more Th17 cells to begin with were better able to control the virus. They then gave animals drugs that deplete Th17 cells and found that reducing the number of Th17 cells made controlling SIV more difficult for those animals.

"We found great variation in the levels of Th17 cells, with as much as a five-fold difference in numbers between animals. e are not sure why this is the case. It could be genetically determined or perhaps due to a previous exposure to a type of bacteria that stimulates production of Th17 cells," said Hartigan-O'Connor.

This study is part of a series of investigations undertaken by researchers at the UCSF Division of Experimental Medicine into how SIV, and by extension HIV, interacts with the immune system in the gut. The previous study was focused on chronic infection and persistent inflammation in the gut.

"The earlier study addressed the cause and consequence of inflammation after infection. We found that inflammation induces an enzyme that knocks out Th17 cells, which normally help to keep the gut intact, and that disease progression was faster. Reciprocally, we have now found that animals do better if they have many Th17 cells at the outset of infection. We are gradually increasing our understanding of this important aspect of the immune system and we are working to translate this understanding into an approach that benefits patients," said study senior author, Joseph M. McCune, MD, PhD, chief of the UCSF Division of Experimental Medicine.

Study co-investigators include Bittoo Kanwar from UCSF Division of Experimental Medicine and Kristina Abel and Koen K. A. Van Rompay from the University of California, Davis.

Insurance superintendent approves Blue Cross rate increase ...

State Superintendent of Insurance John G. Franchini has approved a 6.9 percent rate increase for Blue Cross Blue Shield of New Mexico that will impact about 31,000 of the company?s policyholders in its open and closed blocks of business starting June 1.

Franchini previously denied BCBSNM?s request for a 9.9 percent rate increase, instead opting for the lower percentage which more closely matches current medical cost inflation.

In its filing, BCBSNM asked for higher rates to help cover the cost of supplying policies to individual plan members in rural parts of the state where medical costs are more expensive. According to the company, about 70 percent of its individual plan members live outside the Albuquerque metropolitan area.

?This was a very thorough rate review process that will have a beneficial outcome for thousands of people throughout the state who are in dire need of access to affordable healthcare,? Franchini said.

John Shore: Director of Americans United for Separation of Church and State writes IRS about Pastor Charles Worley

Barry W. Lynn, executive director of Americans United for Separation of Church and State, has written a letter to Lois G. Lerner, director of the IRS's Exempt Organizations Division, in which he requests that Ms. Lerner consider that in this sermon Pastor Charles Worley violated federal law by intervening in an election.

Dated May 23, 2012, Mr. Lynn's letter reads:

I am writing today with information about a church in Maiden, N.C., that I believe has violated
federal law by intervening in an election.

On May 13, Pastor Charles L. Worley of Providence Road Baptist Church delivered a sermon denouncing President Barack Obama's support for same-sex marriage.

During his remarks, Worley suggested quarantining gays and lesbians and allowing them to die. These hateful and repugnant statements attracted much media attention. (See news story enclosed.) But it should not be overlooked that Worley's comments also included a partisan appeal related to the November election.

Worley referred to "our president getting up and saying that it was all right for two women to marry or two men to marry" and added, "I was disappointed bad." He then went on to say, "Someone said, 'Who ya gonna vote for?' I ain't gonna vote for a baby killer and a homosexual lover. You said, 'Did you mean to say that?' You better believe I did."

In context, it is clear that Worley is urging congregants to vote against Obama in the presidential election.

As you know, federal tax law prohibits churches and other 501(c)(3) nonprofits from intervening in elections on behalf of or in opposition to any candidate for public office. When the top official of a religious organization - the chief pastor of the church - issues an appeal to congregants from the pulpit during a worship service in the strongest possible terms to vote against a candidate, it is clearly intervention in an election.

Church leaders seem to realize that the sermon is problematic. It has been removed from the church's website, and in fact the church's entire website is (as of today) no longer online.

I think Mr. Lynn's letter could prove to be the snowball that finally starts the avalanche that ultimately wipes out political proselytizing from the pulpit of American churches.

From Section 501 of the Internal Revenue Service Tax Code (which is the part that applies to churches):

Organizations that are exempt from in?come tax under section 501(a) of the In?ternal Revenue Code as organizations described in section 501(c)(3) may not par?ticipate in, or intervene in (including the publishing or distributing of statements), any political campaign on behalf of (or in opposition to) any candidate for public of?fice.

...

Section 1.501(c)(3)-1(c)(3)(i) of the In?come Tax Regulations states that an organization is not operated exclusively for one or more exempt purposes if it is an "action" organization.

Section 1.501(c)(3)-1(c)(3)(iii) of the regulations defines an "action" organiza?tion as an organization that participates or intervenes, directly or indirectly, in any po? litical campaign on behalf of or in opposi?tion to any candidate for public office. ... The reg?ulations further provide that activities that constitute participation or intervention in a political campaign on behalf of or in op?position to a candidate include, but are not limited to, the publication or distribution of written statements or the making of oral statements on behalf of or in opposition to such a candidate.

For their or?ganizations to remain tax exempt under section 501(c)(3), leaders cannot make partisan comments in official organization publications or at official functions of the organization.

...

Section 501(c)(3) organizations must avoid any issue advocacy that functions as political campaign intervention. Even if a statement does not expressly tell an audience to vote for or against a specific candidate, an organization delivering the statement is at risk of violating the polit?ical campaign intervention prohibition if there is any message favoring or opposing a candidate. A statement can identify a candidate not only by stating the candi?date's name but also by other means such as showing a picture of the candidate, referring to political party affiliations, or other distinctive features of a candidate' s platform or biography. All the facts and circumstances need to be considered to determine if the advocacy is political campaign intervention.

Key factors in determining whether a communication results in political cam?paign intervention include the following:

? Whether the statement identifies one or more candidates for a given public office;

? Whether the statement expresses ap?proval or disapproval for one or more candidates' positions and/or actions;

? Whether the statement is delivered close in time to the election;

? Whether the statement makes refer?ence to voting or an election;

? Whether the issue addressed in the communication has been raised as an issue distinguishing candidates for a given office;

? Whether the communication is part of an ongoing series of communications by the organization on the same issue that are made independent of the tim?ing of any election; and

? Whether the timing of the communi?cation and identification of the candi? date are related to a non-electoral event such as a scheduled vote on specific legislation by an officeholder who also happens to be a candidate for public of?fice.

A communication is particularly at risk of political campaign intervention when it makes reference to candidates or voting in a specific upcoming election.

Seagrasses can store as much carbon as forests

ScienceDaily (May 23, 2012) ? Seagrasses are a vital part of the solution to climate change and, per unit area, seagrass meadows can store up to twice as much carbon as the world's temperate and tropical forests.

So report researchers publishing a paper this week in the journal Nature Geoscience.

The paper, "Seagrass Ecosystems as a Globally Significant Carbon Stock," is the first global analysis of carbon stored in seagrasses.

The results demonstrate that coastal seagrass beds store up to 83,000 metric tons of carbon per square kilometer, mostly in the soils beneath them.

As a comparison, a typical terrestrial forest stores about 30,000 metric tons per square kilometer, most of which is in the form of wood.

The research also estimates that, although seagrass meadows occupy less than 0.2 percent of the world's oceans, they are responsible for more than 10 percent of all carbon buried annually in the sea.

"Seagrasses only take up a small percentage of global coastal area, but this assessment shows that they're a dynamic ecosystem for carbon transformation," said James Fourqurean, the lead author of the paper and a scientist at Florida International University and the National Science Foundation's (NSF) Florida Coastal Everglades Long-Term Ecological Research (LTER) site.

The Florida Coastal Everglades LTER site is one of 26 such NSF LTER sites around the world in ecosystems from forests to tundra, coral reefs to barrier islands.

"Seagrasses have the unique ability to continue to store carbon in their roots and soil in coastal seas," said Fourqurean. "We found places where seagrass beds have been storing carbon for thousands of years."

The research was led by Fourqurean in partnership with scientists at the Spanish High Council for Scientific Investigation, the Oceans Institute at the University of Western Australia, Bangor University in the United Kingdom, the University of Southern Denmark, the Hellenic Center for Marine Research in Greece, Aarhus University in Denmark and the University of Virginia.

Seagrass meadows, the researchers found, store ninety percent of their carbon in the soil--and continue to build on it for centuries.

In the Mediterranean, the geographic region with the greatest concentration of carbon found in the study, seagrass meadows store carbon in deposits many meters deep.

Seagrasses are among the world's most threatened ecosystems. Some 29 percent of all historic seagrass meadows have been destroyed, mainly due to dredging and degradation of water quality. At least 1.5 percent of Earth's seagrass meadows are lost every year.

The study estimates that emissions from destruction of seagrass meadows can potentially emit up to 25 percent as much carbon as those from terrestrial deforestation.

"One remarkable thing about seagrass meadows is that, if restored, they can effectively and rapidly sequester carbon and reestablish lost carbon sinks," said paper co-author Karen McGlathery, a scientist at the University of Virginia and NSF's Virginia Coast Reserve LTER site.

The Virginia Coast Reserve and Florida Coastal Everglades LTER sites are known for their extensive seagrass beds.

Seagrasses have long been recognized for their many ecosystem benefits: they filter sediment from the oceans; protect coastlines against floods and storms; and serve as habitats for fish and other marine life.

The new results, say the scientists, emphasize that conserving and restoring seagrass meadows may reduce greenhouse gas emissions and increase carbon stores--while delivering important "ecosystem services" to coastal communities.

The research is part of the Blue Carbon Initiative, a collaborative effort of Conservation International, the International Union for Conservation of Nature, and the Intergovernmental Oceanographic Commission of UNESCO.

Delay of bloom blamed on climate change

Flowers that fail to open early in spring may be taking cue from previous autumn

Oddball spring flowers that seem unaffected by climate change ? or have gotten it backwards ? could be taking some cues from unusual autumns the year before.

Like their early flowering peers, plants in a new study show that many of these on-time and late bloomers also have had their timing shifted by a warming globe ? but the paradoxical effects have just been more difficult to observe.

Climate change is bringing warmer and earlier springtimes to meadows, mountain slopes and many other ecosystems worldwide, with many a bud bursting into bloom days or weeks earlier than in decades past. Yet a minority of plants continue to bloom at their regular time, and some even appear to delay their blooms, says Benjamin Cook of the NASA Goddard Institute for Space Studies in New York City. While some have suggested explanations for these contrarian plants? out-of-step timing, the effect has been tricky to analyze.

Looking at plant bloom times in terms of both spring and fall temperature quirks, however, shows that some species are sensitive to signals from unusual fall temperatures, Cook and his colleagues report online May 21 in Proceedings of the National Academy of Sciences. Warmer fall temperatures may mask or delay any tendencies to bloom earlier in the spring.

Biologists have known that certain plants need a good chill over the winter before they thrill to the warming of spring and break out of dormancy to bloom. If weather doesn?t cool down as soon as it used to in the fall, then the chilling doesn?t start on time and there?s a delay in the spring response. Requirements for this cool-down time-out, called vernalization, are better known for crops and garden beauties than for most wild species.

To try to quantify how climate change tweaks vernalization for a wide range of species, Cook and his colleagues turned to a 47-year record (1954 through 2000) of the first bloom dates for 384 species growing near the same site around Chinnor, England. And to broaden the range, the researchers looked at 31 years of first blooms from 106 species around Washington, D.C.

Statistical methods found that most of the plants ? 72 percent in England and 73 percent in the United States ? responded only to spring warming and bloomed earlier, but showed no discernible influence of warming fall temperatures.

For many of the other plants though, adding in a delay caused by a warm fall made the springtime pattern make sense at last. For example, Clematis vitalba, a relative of the vines common in gardens, at first glance doesn?t appear affected by climate change. Yet the analysis showed a tendency to advance bloom 5.1 days in spring, which a delaying pull from the previous fall cancels.

In contrast Chinnor records show the ground-covering bugleweed (Ajuga reptans) blooming almost six days later than it used to. Warming springs are urging the bloom time forward, the new analysis shows, but not strongly enough to compensate for a big delay from autumn.

Although vernalization is an old concept, ?What is new here is that they believe you can be predictive,? says ecologist Alastair Fitter of the University of York in England. He and his father had collected the Chinnor data, but he wasn?t part of the new analysis.

Understanding what?s going on with the species that don?t seem to have gotten the memo on climate change could have practical value for conservationists, he says. Decisions about management could turn out quite differently if biologists figure out that plants involved are responding to fall as well as spring climate glitches.

Obama sits down for exclusive interview with ABC

(REUTERS/Larry Downing)President Obama will sit down with "Good Morning America" anchor Robin Roberts today at the White House for a wide-ranging interview, his first since Vice President Joe Biden's public support of same-sex marriage and North Carolina voters imposed a new ban on all same-sex unions. When asked Monday about the president's self-described evolution on the issue, White House Press Secretary Jay Carney said, "I can tell you that I'm sure it is the case that he will be asked again at some point when he gives interviews or press conferences about this issue, and I'll leave it to him to describe his personal views."

With his "evolving" stance on same-sex marriage in the spotlight, here's a look back at the various positions he has held on the issue: from appearing to support the unions as a young state senate candidate, opposing them outright as a matter of faith in 2004, to suggesting a shift in line with public opinion:

FEBRUARY 1996:??? "I favor legalizing same-sex marriages, and would fight efforts to prohibit such marriages," reads a typed, signed statement from then-Illinois state senate candidate Obama in response to a questionnaire by the Chicago LGBT newspaper "Outlines." ??White House Communications Director Dan Pfeiffer later publicly disavowed the statement, claiming in June 2011 that the questionnaire was "actually filled out by someone else."

OCTOBER 2004:? " What I believe is that marriage is between a man and a woman ? What I believe, in my faith, is that a man and a woman, when they get married, are performing something before God, and it's not simply the two persons who are meeting," then-U.S. Senate candidate Obama said in an interview with WTTW Chicago public television.

"That doesn't mean that that necessarily translates into a position on public policy or with respect to civil unions. What it does mean is that we have a set of traditions in place that, I think, need to be preserved, but I also think we need to make sure that gays and lesbians have the same set of basic rights that are in place.

"I don't think marriage is a civil right," Obama said when asked whether there's an inherent right to marry.

OCTOBER 2010:?? " I have been to this point unwilling to sign on to same-sex marriage primarily because of my understandings of the traditional definitions of marriage," President Obama said during an interview with liberal bloggers. "But I also think you're right that attitudes evolve, including mine. And I think that it is an issue that I wrestle with and think about because I have a whole host of friends who are in gay partnerships."

DECEMBER 2010:?? " My feelings about this are constantly evolving. I struggle with this. At this point, what I've said is, is that my baseline is a strong civil union that provides them the protections and the legal rights that married couples have," Obama said in response to a question from ABC's Jake Tapper at a White House press conference.

"I recognize that from their perspective it is not enough, and I think is something that we're going to continue to debate and I personally am going to continue to wrestle with going forward," he said.

JUNE 2011:? " The president has never favored same-sex marriage. He is against it. The country is evolving on this, and he is evolving on it," Pfeiffer told progressive activists at the Net Roots Nation conference.

JUNE 2011:? " I think it's important for us to work through these issues because each community is going to be different, each state is going to be different," Obama said when asked during a White House press conference about New York becoming the latest state to legalize same-sex marriage.

"I think what you're seeing is a profound recognition on the part of the American people that gays and lesbians and transgender persons are our brothers, our sisters, our children, our cousins, our friends, our co-workers, and that they've got to be treated like every other American," he said. "And I think that principle will win out. It's not going to be perfectly smooth, and it turns out that the President - I've discovered since I've been in this office - can't dictate precisely how this process moves."

OCTOBER 2011:? "I'm still working on it," Obama said when asked by ABC's George Stephanopoulos whether he would move from supporting civil unions for same-sex couples to supporting gay marriage.

"I probably won't make news right now, George. But I think that there's no doubt that as I see friends, families children of gay couples who are thriving, you know, that has an impact on how I think about these issues."
